Abstract: To study the application of DNA barcoding in bryophytes community, bryophytes within the sample area were collected across-the-board based on Baiyun Mountain National Forest Park four 1 hm~2(100 m×100 m) forest dynamic monitoring plots. Three gene fragments of Rps4, rbcl and trnL-F were used for single and combined DNA sequencing.Then the application success rate of DNA barcoding were evaluated in bryophytes, and the phylogenetic evolution relationship tree of bryophytes was constructed based on the DNA sequences. The results showed that: a total of 71 species of bryophytes were collected, belonging to 45 genera and 25 families, including 66 species of mosses in 41 genera of 21 families, 5 species of liverworts in 4 genera of 4 families;the comprehensive success rate of rps4 was the highest(95.81%) for a single DNA barcoding fragment, followed by trnL-F(85.53%) and rbcL(67.8%), and the comprehensive success rate of the combined DNA barcoding fragment were all more than 75.59%. The phylogenetic evolutionary relationship tree constructed by DNA barcoding fragments of bryophytes showed that liverworts and mosses of Baiyun Mountain National Forest Park formed two branches, and the support rate reached a high level of 98 percent.
